ELISA 酶结合物的制备:免疫酶技术(immunoenzymatic technique)又称酶免疫测定法(enzyme immunoassay,EIA),是继免疫荧光技术和放射免疫技术之后发展起来的又一种免疫标记技术。它是把抗原抗体的特异性反应和酶的高效催化作用相结合而建立的。该技术通过化学方法将酶与抗体或抗抗体结合,形成酶标记物,这些酶标记物仍保持免疫学活性和酶的活性,然后将它与相应的抗体或抗原起反应,形成酶标记复合物,结合在免疫复合物上的酶,在遇到相应的底物时,则催化底物产生水解、氧化或还原等反应,而生成可溶性或不溶性有色物质。然后根据显色的深浅来反映待测样品中抗原或抗体的含量,如生成的有色物质为可溶性,则可用肉眼比色或酶标测定仪测定光密度值(OD)定性或定量;如生成的有色物质为不溶性沉淀物,同时又是电子致密物质,则可用光学显微镜或电子显微镜识别和定位。因此,免疫酶技术是一项定位、定性和定量(敏感度可达每毫升ng~pg水平)的综合技术。常用的酶是辣根过氧化物酶(horseradish peroxidase,HRP)和碱性磷酸酶(alkaline phosphatase,AKP)。
